"One occupant of the car has been taken to a London hospital and the other two have been taken to a local hospital.
"Motorists are advised to expect delays and, where possible, use an alternative route."
Whitfield Hill is understood to be shut between the junctions with the A2 and London Road.
The road re-opened at midday.
Detectives are carrying out an investigation into the circumstances surrounding the incident at the scene.
