In the shocking video posted on social media, a man gets out of a vehicle and walks towards a parked black pick-up truck.
A Dogue de Bordeaux has his head poking out the back window, and the man approaches the dog before slapping him around the face.
A second dog of the same breed then emerges through the open window and barks at the man who retreats while he - and the person filming the video - laugh.
RSPCA investigators believe the video was filmed in the UK - due to the number plates of passing vehicles - but it’s unclear where the incident occurred.
Chief inspector at the charity, Tony Pritchard, said: “We’re keen to hear from the owner of these dogs. They may be completely unaware of this incident.
“We’d like to check that this dog is okay and gather some more information about what happened here.
“We’re also appealing to the public to get in touch with us if they recognise the culprit in the video.”
The RSPCA has had six calls relating to the appeal as of November 1 and has thanked these people for their help.
A spokesman said: "We would like to thank the kind members of the public who have contacted us following the appeal.
"This is an on-going investigation so at this stage we cannot provide any further information."
The man who hit the dog is described as white with fair hair. He was wearing black shiny shoes, black trousers and a white, long-sleeve top.
Anyone who believes they recognise the dogs, know where this incident took place or has any information on who is responsible can contact the RSPCA on 0300 123 8018.
